L’Institut de Recherches SERVIER avec le patronage de la Société de Chimie Thérapeutique lance un appel à projets dans le cadre destages post-doctoraux et thèses de doctorats. L’objet de ces financements est de favoriser l’éclosion de jeunes talents en Chimie Thérapeutique et de permettre la réalisation de programmes de recherche de haute valeur dans des équipes de dimension européenne.

Le thème retenu pour cet appel à projets 2018 est :

Metabolic remodeling in age related diseases

Aging is the main risk factor for a number of chronic diseases such as diabetes, cardiovascular diseases (heart failure : HF, cardiac hypertrophy), neurodegenerative diseases and cancer. It is well established that all these diseases, and aging itself, are characterized by metabolic remodeling and mitochondrial dysfunction. For example, heart failure and neurodegenerative diseases are characterized by an energetic deficit. More precisely, in HF, maladaptive metabolic remodeling takes different forms across multiple energy-handling domains. Lack of metabolic flexibility and reserve (substrate, oxidative, and phosphotransfer) represents a final common denominator ultimately compromising efficiency and contractile reserve in stressful contexts. Similarly, multiple mechanisms have recently emerged as potential contributors to insulin resistance and type 2 diabetes progression, among them metabolic inflexibility and mitochondrial dysfunction. Indeed, under normal physiological conditions, the organism is able to switch from fat to carbohydrate oxidation according to nutrients availability. Mitochondria through a network of finely tuned metabolic and signaling events orchestrate these transitions in fuel choice. In conditions of insulin resistance, metabolic inflexibility results in a lower glucose oxidation during insulin-stimulated conditions and in a relative low activation of lipid catabolism, which further supports the accumulation of ectopic fat and lipotoxicity with a deleterious effect on insulin signaling.
Different expert consensus statements indicate that restoring these alterations by pharmacological approaches could be a strategy for the treatment of heart failure and insulin resistance in clinical practice. Considerable potential exist to fill this unmet need. In this context, the overall objective of this Servier research proposal is to validate a new Drug Discovery program aiming at identifying compounds able to modulate targets involved in metabolic inflexibility and mitochondrial dysfunction. The aim of the project will be to dissect the regulatory nodes involved in mitochondrial dysfunction and metabolic reprogramming and to propose a medicinal chemistry strategy focusing more particularly in diabetes and cardiovascular diseases. The project will be to propose a new therapeutic solution/modality able to reduce the energetic deficit observed in the aforementioned diseases.

Les laboratoires candidats transmettront par courrier postal (en un seul exemplaire) avant le 15 Juin 2018 minuit au secrétariat de la SCT un dossier de candidature exposant de manière synthétique et non confidentielle les recherches envisagées.
